IONS FOR USE Preparation of Wash Buffer If crystals are present warm the concentrate to dissolve For each microplate add 50mL PBS Tween concentrate MAPT to 950mL of distilled water Label the bottle WASH BUFFER Store at 2 8 C Preparation of Samples Collect patient blood by standard venipuncture procedure using an anticoagulant Lyse blood by freezing use the lysed blood as the test specimen SAMPLE Serum or plasma may be used as an alternative to whole blood but the use of these samples may result in the loss of sensitivity The blood should be stored below 10 C if the analysis is delayed Positive and Negative Controls For Standard and Bulk versions of the kit use the Conjugate Diluent MACD undiluted as the kit Negative Control and use a confirmed Malaria positive blood sample as the Positive Control Assay Procedure Bring all reagents to room temperature 18 25 C before use Prepare WASH BUFFER see Preparation of Wash Buffer Remove required number of MAMW strips Reseal the foil bag containing unused microwell strips immediately with tape Pipette 100uL of the SAMPLE positive control MAPC or known positive and negative control MANC or MAC D into individual microwells Include two positive and two negatives in each assay run Cover and incubate for one 1 hour at room temperature in a humid chamber In the last 10 minutes of the incubation period prepare the working strength CONJUGATE Add 5uL of conjugate

Positivkontrolle eine als positiv best tigte Patientenprobe verwenden Testdurchf hrung Vor Gebrauch alle Reagenzien auf Raumtemperatur bringen WASH BUFFER vorbereiten siehe oben Die entsprechende Anzahl an MAMM Streifen herausnehmen Danach die Verpackung mit unben tzten Mikrotiter Streifen sofort wieder mit Klebeband verschlie en 100uL der SAMPLE Positivkontrolle MAPC und Negativkontrolle IMANC oder MACD in die einzelnen Vertiefungen pipettieren Es sollten bei jedem Testansatz je 2 Kontrollen verwendet werden Anschlie end die Sreifen abdecken und in der feuchten Kammer f r 1 Stunde bei Raumtemperatur inkubieren In den letzten 10 Minuten der Inkubationszeit das CONJUGATE vorbereiten 5uL des Konjugat Konzentrat MAPOJ zu 995uL des Konjugat Verd nnungsmediums MACD geben und gut mischen pro Streifen mit 8 Vertiefungen 1mL L sung herstellen Die Streifen bevorzugt mit einem Waschautomaten waschen ansonsten per Hand Den Inhalt der Vertiefungen ausleeren und WASH BUFFER hinzugeben Das Ganze 4 mal wiederholen Nach dem f nften Mal restliche Tropfen durch Klopfen der umgekehrten Platte auf einem Papierhandtuch entfernen Vorsicht Streifen k nnen aus der Halterung herausfallen 100uL CONJUGATE in jede Vertiefung pipettieren Anschlie end die Sreifen abdecken und in der feuchten Kammer f r 1 Stunden bei Raumtemperatur inkubieren In den letzten 10 Minuten der Inkubationszeit das SUBSTRATE vorbereiten 50uL
